The following: disproved 2015-07-19 after visit to Mudgee Family History Group.
Using the Emigration records held by NSW State Archives regarding 1841 arrival of Ship RUNNYMEDE - Edward Comber's parents are shown as Thomas COMER and Bridget (Unknown) COMER.

These COMERs are NOT the same family as married Elizabeth Mackey.

Also, email correspondence with East Galway Family History Society on 6 Feb shows the bapism of Edward Comer:
A cursory examination of our database of church birth/baptism records did reveal a Edmund Comer born in 1820 in the Roman Catholic parish of Tynagh, but as we have no details of his parent’s name I am unable to determine how/if this record could be relevant to your ancestry. An examination of our church marriage record failed to note any details pertaining to Edward Comer, or variation and where a wife first name appears as Ann, or variation.

Each entry records the progressive number and date of the application, name and address of the applicant, type of employment offered, and result of the application.
The volumes are of the same kind used to register and index ordinary letters received. These applications were processed separately for administrative convenience
